Olympus/Evident VS200 Slide scanner
The Olympus/Evident VS200 is an automated whole-slide imaging system for high-resolution digitization of tissue sections in brightfield and multichannel fluorescence. The system accommodates up to six standard microscope slides per scanning run and supports imaging from low-magnification whole-section overview scans to high-resolution 40× dry and oil-immersion acquisition.
Capabilities
- Automated whole-slide scanning in brightfield and fluorescence
- Up to 6 standard slides per scanning run
- Objectives from 2× to 40×, including high-NA 40× dry (NA 0.95) and 40× oil immersion (NA 1.4)
- Automated oil dispensing for unattended high-resolution oil-immersion scanning
- Multichannel fluorescence, Z-stack and multiplex acquisition
- Sensitive fluorescence detection with a Hamamatsu ORCA-Fusion CMOS camera
- Fluorescence configuration suitable for DAPI, CFP, FITC/Alexa Fluor 488, Cy3, TxRed/mCherry, Cy5 and Cy7
- VSI datasets can be exported to TIFF/BigTIFF, JPEG/JPEG2000 and PNG
Objectives
| Objective | NA | Application |
|---|---|---|
| 2× PLN | 0.06 | Label scan / Rapid overview |
| 4× UPLFLN | 0.13 | Whole-section overview |
| 10× UPLXAPO | 0.40 | Routine whole-slide imaging |
| 20× UPLXAPO | 0.80 | High-resolution whole-slide imaging |
| 40× UPLXAPO dry | 0.95 | High-resolution ROI-scanning |
| 40× UPLXAPO oil | 1.40 | Maximum-resolution ROI-scanning |
Fluorescence imaging
- Fluorescence acquisition uses an LED-based X-Cite NOVEM-L / XT920 illumination system and a Hamamatsu ORCA-Fusion CMOS camera with 2304 × 2304 pixels, 6.5 µm pixel size and a specified quantum efficiency of 80% at 550 nm.
- Installed filter: Fast Pentaband wheel: DAPI • FITC/Alexa 488 • Cy3 • Cy5 • Cy7; Extra cubes: CFP• TxRed/mCherry.
Compatibility with a specific fluorophore should be checked against its excitation and emission spectrum when planning an experiment.
Software
- VS-ASW acquisition software: OLYMPUS VS200 ASW 4.2 (Build 31689).
- VSI viewing software: OlyVIA is available free-of-charge from the manufacturer website.
Typical applications
- Whole-slide digitization of histological specimens
- large-area fluorescence imaging of histological specimens
- multichannel neuroanatomical mapping
- screening and documentation of serial sections
- high-resolution imaging of selected tissue regions
- quantitative downstream image-analysis workflows
Sample Methods Section
“Widefield fluorescence images were acquired using an Olympus VS200 slide-scanning microscope (Evident Scientific/Olympus, Hamburg, Germany), equipped with an X-Cite NOVEM-L LED illumination system (Excelitas Technologies, Waltham, MA, USA)) and a Hamamatsu ORCA-Fusion CMOS camera (Hamamatsu Photonics, Hamamatsu City, Japan). Images were acquired using a [10x/0.40 / 20x/0.80 / 40x/0.95 dry / 40x/1.40 oil] objective and Olympus VS200 ASW V4.2 acquisition software. [Single optical planes / Z-stacks with a z-step size of … µm / Extended focal images of the … µm thick sections] were acquired in the [DAPI/CFP/FITC/Cy3/TxRed/Cy5/Cy7] channel(s). Where required, adjacent image fields were automatically acquired and stitched to generate [whole-section / large-area] mosaic images.”
